Expression analysis of β-secretase (BACE1) and its naturally occurring anti-sense (BACE1-AS) in multiple sclerosis

The β-site APP-cleaving enzyme 1 (BACE1) is the principal enzyme in the cleavage of amyloid precursor protein. It participates in the pathogenesis of neurologic disorders such as Alzheimer's disease and multiple sclerosis (MS). More recently, the naturally occurring antisense for this transcript (BACE1-AS) has been shown to stabilize the expression and function of BACE1. In the present study, we assessed expression of BACE1 and BACE1-AS in peripheral blood of 50 MS patients who were responsive to IFNβ-1a treatment and 50 healthy subjects by means of real time PCR. We found neither significant difference in their expression levels between MS patients and healthy subjects nor any correlations between their expression levels and patients' characteristics. However, we detected a significant correlation between transcript levels of BACE1 and BACE1-AS both in patients and healthy subjects which is in line with the proposed role for BACE1-AS in stabilization of BACE1. The similar expression patterns of BACE1 and BACE1-AS in MS patients and healthy subjects might be due to the effect of IFNβ-1a treatment on their expression. So, future studies are needed to elaborate the underlying mechanism of this observation.
