The effect of pentoxifylline on malathion-induced mitochondrial damage in rat liver

چکیده
Objective: Malathion is one of the most toxic organophosphorus insecticides (OPIs).OPIs may induce oxidative stress in various tissues leading to generation of free radicals and alteration in antioxidant system. The objective of this study was to investigate the possible protective effects ofpentoxifylline as a phosphodiesterase-5 inhibitor on malathion-induced oxidative damage to rat liver mitochondria. Methods: Rats received malathion (200 mg/kg/ day) and pentoxifylline (PTX, 50 mg/kg/day) in combination or alone. After two week of treatment, whole liver tissue, and liver mitochondria were isolated. The activity of enzymatic scavengerssuch as catalase (CAT) and superoxide dismutase (SOD) were measured. The extents of cellular lipid peroxidation (LPO) was determined. The viability was determined in liver mitochondria. Results: Malathion stimulated activities of CAT, SOD and increased LPO in whole liver homogenate. The changes in CAT, SOD and LPO, were restored by PTX. In liver mitochondria, malathion enhanced LPO and decreased viability that was recovered by PTX. Conclusion: It is concluded that oxidative damage is the mechanism of toxicity of malathion in the mitochondria that can be recovered by PTX.
